Uncertain significance for Charcot-Marie-Tooth disease type 4 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014845.6(FIG4):c.374A>G (p.Tyr125Cys), citing Invitae Variant Classification Sherloc (09022015): This variant is present in population databases (rs145466148, ExAC 0.001%). This sequence change replaces tyrosine with cysteine at codon 125 of the FIG4 protein (p.Tyr125Cys). The tyrosine residue is highly conserved and there is a large physicochemical difference between tyrosine and cysteine. This variant has not been reported in the literature in individuals with FIG4-related disease. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of sequence changes on RNA splicing suggest that this variant may create or strengthen a splice site, but this prediction has not been confirmed by published transcriptional studies.
