NM_004656.4(BAP1):c.272G>A (p.Cys91Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.C91Y variant (also known as c.272G>A), located in coding exon 5 of the BAP1 gene, results from a G to A substitution at nucleotide position 272. The cysteine at codon 91 is replaced by tyrosine, an amino acid with highly dissimilar properties. This alteration was non-functional in a high throughput genome editing haploid cell survival functional assay (Waters AJ et al. Nat Genet, 2024 Jul;56:1434-1445). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
