NM_001399.5(EDA):c.641T>A (p.Met214Lys) was classified as Likely pathogenic for Hypohidrotic X-linked ectodermal dysplasia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces methionine with lysine at codon 214 of the EDA protein (p.Met214Lys). The methionine residue is highly conserved and there is a moderate physicochemical difference between methionine and lysine. This variant is not present in population databases (ExAC no frequency). This variant has been observed to segregate with ectodermal dysplasia in a family (Invitae).
