NM_006231.4(POLE):c.4894A>G (p.Met1632Val) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.M1632V variant (also known as c.4894A>G), located in coding exon 37 of the POLE gene, results from an A to G substitution at nucleotide position 4894. The methionine at codon 1632 is replaced by valine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
