NM_000388.4(CASR):c.28C>A (p.Leu10Ile) was classified as Uncertain significance for Nephrolithiasis/nephrocalcinosis by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.L10I variant (also known as c.28C>A), located in coding exon 1 of the CASR gene, results from a C to A substitution at nucleotide position 28. The leucine at codon 10 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
