The valine at codon 515 is replaced by methionine, an amino acid with highly similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the supporting evidence, this variant is unlikely to be causative of Charcot-Marie Tooth type 2D (CMT2D) and distal hereditary motor neuronopathy type VA; however, its contribution to the development of GARS-related mitochondrial respiratory chain dysfunction is uncertain.
