NM_000393.5(COL5A2):c.379A>C (p.Ile127Leu) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.I127L variant (also known as c.379A>C), located in coding exon 5 of the COL5A2 gene, results from an A to C substitution at nucleotide position 379. The isoleucine at codon 127 is replaced by leucine, an amino acid with highly similar properties. This variant has been reported in subject with hypermobility and aortic root dilation (Rashed ER et al. Vasc Med, 2022 Jun;27:283-289). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
