Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_002474.3(MYH11):c.3149G>A (p.Arg1050Gln), citing Ambry Variant Classification Scheme 2023: The p.R1050Q variant (also known as c.3149G>A), located in coding exon 24 of the MYH11 gene, results from a G to A substitution at nucleotide position 3149. The arginine at codon 1050 is replaced by glutamine, an amino acid with highly similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
