This variant lies in the COL3A1 gene (transcript NM_000090.4) at coding-DNA position 1684, where C is replaced by T; at the protein level this means converts the codon for arginine at residue 562 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This variant changes 1 nucleotide in exon 24 of the COL3A1 gene, creating a premature translation stop signal. This variant is expected to result in an absent or non-functional protein product. To our knowledge, this variant has not been reported in individuals affected with cardiovascular disorders in the literature. This variant has not been identified in the general population by the Genome Aggregation Database (gnomAD). Loss of COL3A1 function is a known mechanism of disease (clinicalgenome.org). Based on the available evidence, this variant is classified as Pathogenic.
