Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000264.5(PTCH1):c.121C>A (p.Arg41Ser), citing Ambry Variant Classification Scheme 2023: The p.R41S variant (also known as c.121C>A), located in coding exon 1 of the PTCH1 gene, results from a C to A substitution at nucleotide position 121. The arginine at codon 41 is replaced by serine, an amino acid with dissimilar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this alteration remains unclear.
