Uncertain significance for Primary ciliary dyskinesia — the classification assigned by Ambry Genetics to NM_178452.6(DNAAF1):c.1318G>A (p.Gly440Arg), citing Ambry Variant Classification Scheme 2023: The p.G440R variant (also known as c.1318G>A), located in coding exon 8 of the DNAAF1 gene, results from a G to A substitution at nucleotide position 1318. The glycine at codon 440 is replaced by arginine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
