This variant lies in the TTN gene (transcript NM_001267550.2) at coding-DNA position 50441, where C is replaced by G; at the protein level this means replaces threonine at residue 16814 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are unavailable for the TTN gene. This variant is not present in population databases (ExAC no frequency). This sequence change replaces threonine with serine at codon 16814 of the TTN protein (p.Thr16814Ser). The threonine residue is highly conserved and there is a small physicochemical difference between threonine and serine. This variant has not been reported in the literature in individuals with TTN-related disease. This variant is located in the A band of TTN (PMID: 25589632). Variants in this region may be relevant for cardiac or neuromuscular disorders (PMID: 25589632, 23975875).
