NM_006231.4(POLE):c.5293T>C (p.Ser1765Pro) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S1765P variant (also known as c.5293T>C), located in coding exon 39 of the POLE gene, results from a T to C substitution at nucleotide position 5293. The serine at codon 1765 is replaced by proline, an amino acid with similar properties. This amino acid position is conserved. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
