This variant lies in the DST gene (transcript NM_001723.7) at coding-DNA position 6415, where A is replaced by G; at the protein level this means replaces isoleucine at residue 2139 with valine — a missense variant. Submitter rationale: This sequence change replaces isoleucine with valine at codon 2139 of the DST protein (p.Ile2139Val). The isoleucine residue is weakly conserved and there is a small physicochemical difference between isoleucine and valine. This variant is present in population databases (rs374067079, ExAC 0.003%). This variant has not been reported in the literature in individuals affected with DST-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
