NM_000243.3(MEFV):c.766A>G (p.Asn256Asp) was classified as Uncertain significance for Familial Mediterranean fever by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces asparagine with aspartic acid at codon 256 of the MEFV protein (p.Asn256Asp). The asparagine residue is moderately conserved and there is a small physicochemical difference between asparagine and aspartic acid. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Tolerated"; PolyPhen-2: "Possibly Damaging"; Align-GVGD: "Class C0"). This variant has not been reported in the literature in individuals with MEFV-related disease. This variant is not present in population databases (ExAC no frequency).
