Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_001148.6(ANK2):c.3068G>A (p.Gly1023Glu), citing Ambry Variant Classification Scheme 2023. This variant lies in the ANK2 gene (transcript NM_001148.6) at coding-DNA position 3068, where G is replaced by A; at the protein level this means replaces glycine at residue 1023 with glutamic acid — a missense variant. Submitter rationale: The p.G1023E variant (also known as c.3068G>A), located in coding exon 27 of the ANK2 gene, results from a G to A substitution at nucleotide position 3068. The glycine at codon 1023 is replaced by glutamic acid, an amino acid with similar properties. This variant was not reported in population based cohorts in the following databases: Database of Single Nucleotide Polymorphisms (dbSNP), NHLBI Exome Sequencing Project (ESP), and 1000 Genomes Project. In the ESP, this variant was not observed in 6503 samples (13006 alleles) with coverage at this position. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
