This variant lies in the ATL1 gene (transcript NM_015915.5) at coding-DNA position 572, where A is replaced by G; at the protein level this means replaces glutamine at residue 191 with arginine — a missense variant. Submitter rationale: This sequence change replaces glutamine, which is neutral and polar, with arginine, which is basic and polar, at codon 191 of the ATL1 protein (p.Gln191Arg). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with autosomal dominant hereditary spastic paraplegia (PMID: 17502470; Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 576063).
