This variant lies in the CACNA1A gene (transcript NM_001127222.2) at coding-DNA position 3926, where G is replaced by A; at the protein level this means replaces arginine at residue 1309 with histidine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. This sequence change replaces arginine with histidine at codon 1310 of the CACNA1A protein (p.Arg1310His).
