NM_000138.5(FBN1):c.6595G>A (p.Gly2199Ser) was classified as Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection by Color Diagnostics, LLC DBA Color Health, citing ACMG Guidelines, 2015: This missense variant replaces glycine with serine at codon 2199 of the FBN1 protein. Computational prediction suggests that this variant may have deleterious impact on protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual affected with thoracic aortic aneurysm (Gillis 2019, doctoral thesis at the University of Antwerp, Belgium) and in an individual suspected of having Marfan syndrome, who also carried a variant of uncertain significance in the FBN2 gene (PMID: 31506931). This variant has been identified in 3/251244 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
