NM_025216.3(WNT10A):c.212A>T (p.Gln71Leu) was classified as Uncertain significance for Tooth agenesis, selective, 4; Odonto-onycho-dermal dysplasia by Labcorp Genetics (formerly Invitae), Labcorp, citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces glutamine with leucine at codon 71 of the WNT10A protein (p.Gln71Leu). The glutamine residue is highly conserved and there is a moderate physicochemical difference between glutamine and leucine. This variant is present in population databases (rs368943130, ExAC 0.004%). This variant has not been reported in the literature in individuals affected with WNT10A-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
