Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_000222.3(KIT):c.89G>T (p.Ser30Ile), citing Ambry Variant Classification Scheme 2023: The p.S30I variant (also known as c.89G>T), located in coding exon 2 of the KIT gene, results from a G to T substitution at nucleotide position 89. The serine at codon 30 is replaced by isoleucine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
