Uncertain significance for Familial thoracic aortic aneurysm and aortic dissection — the classification assigned by Ambry Genetics to NM_053025.4(MYLK):c.2305G>A (p.Glu769Lys), citing Ambry Variant Classification Scheme 2023: The p.E769K variant (also known as c.2305G>A), located in coding exon 13 of the MYLK gene, results from a G to A substitution at nucleotide position 2305. The glutamic acid at codon 769 is replaced by lysine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
