NM_006231.4(POLE):c.6173A>G (p.Glu2058Gly) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E2058G variant (also known as c.6173A>G), located in coding exon 45 of the POLE gene, results from an A to G substitution at nucleotide position 6173. The glutamic acid at codon 2058 is replaced by glycine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
