This variant lies in the LRP4 gene (transcript NM_002334.4) at coding-DNA position 3331, where G is replaced by A; at the protein level this means replaces aspartic acid at residue 1111 with asparagine — a missense variant. Submitter rationale: This sequence change replaces aspartic acid with asparagine at codon 1111 of the LRP4 protein (p.Asp1111Asn). The aspartic acid residue is highly conserved and there is a small physicochemical difference between aspartic acid and asparagine. This variant is present in population databases (rs770071302, ExAC 0.001%). This variant has not been reported in the literature in individuals affected with LRP4-related conditions. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be tolerated.
