This variant lies in the BRCA2 gene (transcript NM_000059.4) at coding-DNA position 6067, where G is replaced by T; at the protein level this means replaces aspartic acid at residue 2023 with tyrosine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces aspartic acid with tyrosine at codon 2023 of the BRCA2 protein (p.Asp2023Tyr).
