Uncertain significance for Nemaline myopathy 8 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_152393.4(KLHL40):c.1053C>A (p.His351Gln), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces histidine with glutamine at codon 351 of the KLHL40 protein (p.His351Gln). The histidine residue is highly conserved and there is a small physicochemical difference between histidine and glutamine. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance. Algorithms developed to predict the effect of missense changes on protein structure and function do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C15"). This variant has not been reported in the literature in individuals with KLHL40-related disease. This variant is not present in population databases (ExAC no frequency).
