This variant lies in the MSH6 gene (transcript NM_000179.3) at coding-DNA position 3679, where A is replaced by G; at the protein level this means replaces isoleucine at residue 1227 with valine — a missense variant. Submitter rationale: The p.I1227V variant (also known as c.3679A>G), located in coding exon 8 of the MSH6 gene, results from an A to G substitution at nucleotide position 3679. The isoleucine at codon 1227 is replaced by valine, an amino acid with highly similar properties. This variant was identified in a cohort of 3,579 African males diagnosed with prostate cancer who underwent multi-gene panel testing of 19 DNA repair and cancer predisposition genes (Matejcic M et al.
