Pathogenic for MHC class II deficiency — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_003721.4(RFXANK):c.271_271+1insCTGCC, citing Invitae Variant Classification Sherloc (09022015): For these reasons, this variant has been classified as Pathogenic. Loss-of-function variants in RFXANK are known to be pathogenic (PMID: 10803838, 16166641, 21908431). This variant has not been reported in the literature in individuals with RFXANK-related disease. This variant is not present in population databases (ExAC no frequency). This sequence change creates a premature translational stop signal (p.Leu92Alafs*17) in the RFXANK gene. It is expected to result in an absent or disrupted protein product.
