NM_000143.4(FH):c.974G>C (p.Ser325Thr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S325T variant (also known as c.974G>C), located in coding exon 7 of the FH gene, results from a G to C substitution at nucleotide position 974. The serine at codon 325 is replaced by threonine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
