Pathogenic for Polyglandular autoimmune syndrome, type 1 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_000383.4(AIRE):c.958del (p.Leu320fs), citing Invitae Variant Classification Sherloc (09022015): This sequence change creates a premature translational stop signal (p.Leu320Trpfs*58) in the AIRE gene. It is expected to result in an absent or disrupted protein product. Loss-of-function variants in AIRE are known to be pathogenic (PMID: 11524731, 26141571). This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with autoimmune polyendocrine syndrome type 1 (PMID: 21865375). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 573407). For these reasons, this variant has been classified as Pathogenic.
