NM_006231.4(POLE):c.1037G>A (p.Arg346Lys) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.R346K variant (also known as c.1037G>A), located in coding exon 11 of the POLE gene, results from a G to A substitution at nucleotide position 1037. The arginine at codon 346 is replaced by lysine, an amino acid with highly similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
