This variant lies in the NLRC4 gene (transcript NM_001199138.2) at coding-DNA position 494, where G is replaced by C; at the protein level this means replaces cysteine at residue 165 with serine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain. This sequence change replaces cysteine with serine at codon 165 of the NLRC4 protein (p.Cys165Ser). The cysteine residue is highly conserved and there is a moderate physicochemical difference between cysteine and serine.
