This variant lies in the CSRP3 gene (transcript NM_003476.5) at coding-DNA position 109, where T is replaced by G; at the protein level this means replaces cysteine at residue 37 with glycine — a missense variant. Submitter rationale: This sequence change replaces cysteine with glycine at codon 37 of the CSRP3 protein (p.Cys37Gly). The cysteine residue is highly conserved and there is a large physicochemical difference between cysteine and glycine. This variant is not present in population databases (ExAC no frequency). This variant has not been reported in the literature in individuals with CSRP3-related disease. Algorithms developed to predict the effect of missense changes on protein structure and function (SIFT, PolyPhen-2, Align-GVGD) all suggest that this variant is likely to be disruptive, but these predictions have not been confirmed by published functional studies and their clinical significance is uncertain.
