Submitter rationale: This sequence change replaces asparagine, which is neutral and polar, with serine, which is neutral and polar, at codon 127 of the PRF1 protein (p.Asn127Ser). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with primary hemophagocytic lymphohistiocytosis (PMID: 29357941, 32375849).
