NM_004304.5(ALK):c.3020T>C (p.Phe1007Ser) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.F1007S variant (also known as c.3020T>C), located in coding exon 18 of the ALK gene, results from a T to C substitution at nucleotide position 3020. The phenylalanine at codon 1007 is replaced by serine, an amino acid with highly dissimilar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
