This variant lies in the DST gene (transcript NM_001723.7) at coding-DNA position 6906, where T is replaced by G; at the protein level this means converts the codon for tyrosine at residue 2302 into a premature stop signal — a nonsense variant expected to truncate the protein. Submitter rationale: This sequence change results in a premature translational stop signal in the DST gene (p.Tyr2302*). While this is not anticipated to result in nonsense mediated decay, it is expected to delete the last 348 amino acids of the DST protein. This variant is present in population databases (rs775286129, ExAC 0.01%). This variant has not been reported in the literature in individuals with DST-related disease.
