Pathogenic for Charcot-Marie-Tooth disease type 2 — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_014874.4(MFN2):c.2251C>T (p.Gln751Ter), citing Invitae Variant Classification Sherloc (09022015): This premature translational stop signal has been observed in individual(s) with Charcot-Marie-Tooth disease (PMID: 16714318). In at least one individual the variant was observed to be de novo. This variant is not present in population databases (gnomAD no frequency). This sequence change creates a premature translational stop signal (p.Gln751*) in the MFN2 gene. While this is not anticipated to result in nonsense mediated decay, it is expected to disrupt the last 7 amino acid(s) of the MFN2 protein. ClinVar contains an entry for this variant (Variation ID: 572169). For these reasons, this variant has been classified as Pathogenic.
