Benign — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_001458.5(FLNC):c.4710C>T (p.Gly1570=), citing LabCorp Variant Classification Summary - May 2015: Variant summary: FLNC c.4710C>T alters a conserved nucleotide resulting in a synonymous change. Several computational tools predict a significant impact on normal splicing: Four predict the variant creates a 5' donor site. However, these predictions have yet to be confirmed by functional studies. The variant allele was found at a frequency of 2.4e-05 in 1613262 control chromosomes. The observed variant frequency exceeds the estimated maximal expected allele frequency for disease-causing variants in FLNC. To our knowledge, no occurrence of c.4710C>T in individuals affected with FLNC-related conditions and no experimental evidence demonstrating its impact on protein function have been reported. ClinVar contains an entry for this variant (Variation ID: 572051). Based on the evidence outlined above, the variant was classified as benign.
