Uncertain significance — the classification assigned by Women's Health and Genetics/Laboratory Corporation of America, LabCorp to NM_199242.3(UNC13D):c.1933C>T (p.Arg645Trp), citing LabCorp Variant Classification Summary - May 2015: Variant summary: UNC13D c.1933C>T (p.Arg645Trp) results in a non-conservative amino acid change in the encoded protein sequence. Four of five in-silico tools predict a damaging effect of the variant on protein function. The variant allele was found at a frequency of 4e-05 in 251326 control chromosomes. This frequency is not significantly higher than estimated for a pathogenic variant in UNC13D causing Familial Hemophagocytic Lymphohistiocytosis (4e-05 vs 0.0027), allowing no conclusion about variant significance. c.1933C>T has been reported in the literature in at least one individual with clinical features of Familial Hemophagocytic Lymphohistiocytosis (Vahidi_2019). To our knowledge, no experimental evidence demonstrating an impact on protein function has been reported. The following publication has been ascertained in the context of this evaluation (PMID: 32245292). One submitter has cited clinical-significance assessments for this variant to ClinVar after 2014 and has classified the variant as uncertain significance. Based on the evidence outlined above, the variant was classified as uncertain significance.
