This variant lies in the ALS2 gene (transcript NM_020919.4) at coding-DNA position 4858, where G is replaced by A; at the protein level this means replaces glutamic acid at residue 1620 with lysine — a missense variant. Submitter rationale: Algorithms developed to predict the effect of missense changes on protein structure and function are either unavailable or do not agree on the potential impact of this missense change (SIFT: "Deleterious"; PolyPhen-2: "Probably Damaging"; Align-GVGD: "Class C0"). This sequence change replaces glutamic acid with lysine at codon 1620 of the ALS2 protein (p.Glu1620Lys).
