NM_005585.5(SMAD6):c.173C>G (p.Ser58Cys) was classified as Uncertain significance for Inborn genetic diseases by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S58C variant (also known as c.173C>G), located in coding exon 1 of the SMAD6 gene, results from a C to G substitution at nucleotide position 173. The serine at codon 58 is replaced by cysteine, an amino acid with dissimilar properties. This amino acid position is conserved. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
