NM_177438.3(DICER1):c.1231T>G (p.Ser411Ala) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.S411A variant (also known as c.1231T>G), located in coding exon 7 of the DICER1 gene, results from a T to G substitution at nucleotide position 1231. The serine at codon 411 is replaced by alanine, an amino acid with similar properties. This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
