Uncertain significance for Cardiovascular phenotype — the classification assigned by Ambry Genetics to NM_006939.4(SOS2):c.721G>A (p.Glu241Lys), citing Ambry Variant Classification Scheme 2023: The p.E241K variant (also known as c.721G>A), located in coding exon 6 of the SOS2 gene, results from a G to A substitution at nucleotide position 721. The glutamic acid at codon 241 is replaced by lysine, an amino acid with similar properties. This amino acid position is conserved. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
