Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_007294.4(BRCA1):c.2393C>G (p.Pro798Arg), citing Ambry Variant Classification Scheme 2023: The p.P798R variant (also known as c.2393C>G), located in coding exon 9 of the BRCA1 gene, results from a C to G substitution at nucleotide position 2393. The proline at codon 798 is replaced by arginine, an amino acid with dissimilar properties. This alteration has been previously reported in a Saudi Arabian breast cancer patient (Abulkhair O et al. J Glob Oncol, 2018 08;4:1-9). This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
