Submitter rationale: This sequence change creates a premature translational stop signal (p.Thr34240Hisfs*5) in the TTN gene. While this is not anticipated to result in nonsense mediated decay, it is expected to create a truncated TTN protein. This variant is not present in population databases (gnomAD no frequency). This premature translational stop signal has been observed in individual(s) with dilated cardiomyopathy (PMID: 35653365). This variant is also known as p.Thr34340hisfs*5.
