NM_000179.3(MSH6):c.4051C>T (p.His1351Tyr) was classified as Uncertain significance for Hereditary cancer-predisposing syndrome by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.H1351Y variant (also known as c.4051C>T), located in coding exon 10 of the MSH6 gene, results from a C to T substitution at nucleotide position 4051. The histidine at codon 1351 is replaced by tyrosine, an amino acid with similar properties. This amino acid position is not well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Based on the available evidence, the clinical significance of this variant remains unclear.
