NM_004415.4(DSP):c.3259G>A (p.Glu1087Lys) was classified as Uncertain significance for Cardiovascular phenotype by Ambry Genetics, citing Ambry Variant Classification Scheme 2023: The p.E1087K variant (also known as c.3259G>A), located in coding exon 23 of the DSP gene, results from a G to A substitution at nucleotide position 3259. The glutamic acid at codon 1087 is replaced by lysine, an amino acid with similar properties. This amino acid position is well conserved in available vertebrate species. In addition, the in silico prediction for this alteration is inconclusive. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
