Uncertain significance for Multiple endocrine neoplasia, type 2 — the classification assigned by Color Diagnostics, LLC DBA Color Health to NM_020975.6(RET):c.224C>T (p.Thr75Met), citing ACMG Guidelines, 2015: This missense variant replaces threonine with methionine at codon 75 of the RET protein. Computational prediction suggests that this variant may not impact protein structure and function. To our knowledge, functional studies have not been reported for this variant. This variant has been reported in an individual suspected to be affected with multiple endocrine neoplasia type 2 (doi: 10.5505/aot.2021.25932). This variant has been identified in 10/282298 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
