Pathogenic for Citrullinemia — the classification assigned by Labcorp Genetics (formerly Invitae), Labcorp to NM_054012.4(ASS1):c.1075T>G (p.Tyr359Asp), citing Invitae Variant Classification Sherloc (09022015): This sequence change replaces tyrosine, which is neutral and polar, with aspartic acid, which is acidic and polar, at codon 359 of the ASS1 protein (p.Tyr359Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individuals with citrullinemia (PMID: 19006241; Invitae). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 569667). Advanced modeling of protein sequence and biophysical properties (such as structural, functional, and spatial information, amino acid conservation, physicochemical variation, residue mobility, and thermodynamic stability) performed at Invitae indicates that this missense variant is not expected to disrupt ASS1 protein function with a negative predictive value of 80%. For these reasons, this variant has been classified as Pathogenic.
