Uncertain significance for Hereditary cancer-predisposing syndrome — the classification assigned by Ambry Genetics to NM_032043.3(BRIP1):c.3732G>A (p.Met1244Ile), citing Ambry Variant Classification Scheme 2023: The p.M1244I variant (also known as c.3732G>A), located in coding exon 19 of the BRIP1 gene, results from a G to A substitution at nucleotide position 3732. The methionine at codon 1244 is replaced by isoleucine, an amino acid with highly similar properties. This amino acid position is poorly conserved in available vertebrate species. In addition, this alteration is predicted to be tolerated by in silico analysis. Since supporting evidence is limited at this time, the clinical significance of this alteration remains unclear.
